Active ingredients

Horse chestnut seeds contain 3-6% of a complex mixture of triterpene saponins collectively referred to as aescin (1) , and acylated polyhydroxy oleanene, triterpene oligoglycosides aescins(2) flavonoids eg. Flavonol
glycosides), quinines, sterols, including
stigmasterol , - spinasterol, - sitosterol, fatty acids such as linolenic , palmitic and stearic acids .

Pharmacological actions

As found in different animal experiments,
the principal ingredient in Horse chestnut seeds (HCSE) is the triterpene glycoside mixture, aescin (escin), which has an exclusive vascular –tightening effect. There are indications that HCSE reduces the activity of lysosomal enzymes that is increased in chronic pathological conditions of veins, so that the breakdown of glycolacalyx (mucopolysaccharides ) in the region of the capillary walls is inhibited
.The filtration of low- molecular proteins, electrolytes and water into the interstitium is inhibited through a reduction of vascular permeability . In pharmacological studies involving human subjects and using placebo as reference, a significant reduction of trans- capillary filtration has been demonstrated , and also a remarkable improvement was shown in the symptoms of chronic venous insufficiency(sensation of
tiredness, heaviness and tension , pruritis , pain and swelling in the legs ) in randomized double – blind and cross – over studies .
A randomized placebo – controlled cross over double – blind trial of 22 patients with chronic venous insufficiency concluded that 1200 mg Horse–chestnut extract (standardized to 100 mg Aescin) had an anti edematous effect via a decrease in transcapillary filtration (6) .
